COMPARISON OF TRANSPLATATION OF allograft stem cells INTRAPANCREATIC LAPAROTOMY AND INTRAPERITONIAL TO RENEW PANCREATIC ?-cells ON TYPE 1 DIABETES MELITUS (T1DM) rats

Abstract :

Diabetes mellitus is a degenerative disease that still poses a serious threat to health in Indonesia and the world. Indonesia, ranking fourth after the United States, China, and India, is one of the countries that have the most people with diabetes with the largest population in the world. Alternative therapy using adult stem cells to renew insulin-producing pancreatic ß-cells is a new hope in the prevention of diabetes mellitus type I. Broadly speaking, there are two methods of stem cell transplantation. The first method is to directly implant stem cells into patients’ tissues/organs that are damaged. The second method is to implant stem cells through the blood vessel either the one close to the location of damaged tissues/organs or any blood vessel in the patient's body. Until now, researchers in various stem cell research centers are still trying to find the most optimal method and pathway to administer stem cells into the body. Animals used in this reserch were 18 healthy white male rats Rattus norvegicus Wistar strain aged two to three months with a body weight between 100-150 grams. After adaptation for seven days, the rats were weighed and ten randomly divided into three groups as follows: P0: Animals injected with alloxan with a dose of 150 mg/kgbw intraperitoneally without being given stem cells, P1: Animals injected with alloxan with a dose of 150 mg/kgbw intraperitoneally then given stem cells intraperitoneally, P2: Animals injected with alloxan with a dose of 150 mg/kgbw intraperitoneally and then given stem cells injected intra-pancreatic laparotomy. Observation of HE and immunohistochemical staining was to calculate the mean of pancreatic ?-cells (the number) which is expressed by Pdx-1 and calculated from 3 islets of Langerhans per preparation. Transplantation of allograft pancreatic stem cells using intrapancreatic laparotomy gave better results on ?-cell pancreas renewal in rats with diabetes mellitus type I compared to that using intraperitoneal administration because it is easier to "homing" because it does not require complex preparation phase which usually requires the insertion of genes or other factors that are needed to maximize the administration of stem cells from the site of administration to the damaged area.
